Matthew Chua

Bio

Associate Professor (Dr) Matthew Chua is the winner of the prestigious Leadership Excellence Award 2025 & Global Excellence Award 2024 (SMART Society, USA), as a distinguished leader in the field of AI and technological innovation internationally. He is the Deputy Director of xDigital at the Home Team Science & Technology Agency (HTX), where he leads three enterprise thrusts of Digital Strategy & Plans, Digital Programmes & Engagement, and Digital Transformation to empower frontline missions and transform public sector operations for the future. Matt is also currently an Associate Professor of Biomedical Informatics at NUS School of Medicine. He has also won data science competitions at the national level, clinching 3rd place for the Data VIZard at the Data Arcade Tournament 2022. He has also won the prestigious MINDEF Exemplary Innovator Award for two consecutive years in 2023 and 2024. At NUS, Matt drives the research and lectures on AI and Technology for the advancement of Science and Health. He has won prestigious research grants, amounting to more than S$10 million, from Ministry of Health, SG Enable, and internationally. He has also published more than 50 papers in prestigious peer-reviewed international journals, many of which are in Q1 tier journals, and is widely cited. He is also an Academic Editor for PLOS One Journal.


On the side, Matt is a professional opera singer, being sought after for many local events and celebrations, including The SG50 Concert at the Park, Lee Kuan Yew Memorial Concert, TTSH Gala dinner and many others. He was also the 2nd place winner for The Rising Star Season 3 Competition 2019.

i. Structure‑Gated Protein Intelligence
Dr. Matthew Chua is recognized for pioneering multimodal, structure‑aware protein intelligence frameworks that unify sequence semantics with 3D geometric representations. His early‑fusion architectures, including UniRES‑GO and RIMGOGraph, integrate protein language model embeddings directly with AlphaFold‑derived residue interaction graphs, enabling biologically faithful message passing and significantly improving protein function prediction. His work has helped establish a new methodological standard for structure‑gated protein modeling.

ii. 3D Cross‑Modal Drug Discovery
Dr. Chua’s second core expertise lies in advancing drug–target interaction modeling through massively pre‑trained 3D structural encoders. He developed the 3DICE framework, which jointly leverages Uni‑Mol ligand conformers and ESM‑IF1 protein backbones within an interpretable co‑attention architecture. This approach achieves state‑of‑the‑art performance in cold‑start drug discovery scenarios and demonstrates how geometric encoders can overcome the limitations of traditional 1D sequence‑based models.

iii. Causal Interpretability & Mechanistic Validation
Dr. Chua is internationally known for establishing rigorous causal interpretability standards in biological AI. He introduced deletion‑curve faithfulness tests to verify whether model attention corresponds to true mechanistic residues and atoms, and validated cross‑modal attention weights against crystallographic ground truth using PLIP interaction profiles. His contributions provide the field with principled methods for trustworthy, mechanistically grounded AI in computational biology and drug discovery.
